p38γ

p38γ (MAPK12) is a stress-activated p38 MAPK isoform that controls cellular responses to environmental stress and inflammation[1]. Mechanistically, p38γ/p38δ regulate TLR4-induced cytokine production through ERK1/2 activation, linking this isoform group to innate immune signaling[2]. In skeletal muscle models, p38γ, but not p38α or p38β, is required for endurance exercise-induced mitochondrial biogenesis and angiogenesis[3]. In disease models, p38γ/p38δ deficiency reduces tumor formation, cytokine and chemokine production, and myeloid-cell recruitment in colitis-associated colon cancer[4]. Cancer studies further show that p38γ promotes breast cancer motility through RhoC GTPase and cytoskeletal regulation, supports triple-negative breast cancer stem-like cell expansion, and drives liver tumorigenesis through cell-cycle progression[5][6][7]. Compared with related isoforms, p38α is the best-characterized family member, whereas p38γ shows tissue-enriched activity and a distinctive PDZ-binding motif that supports isoform-specific signaling[1][8]. For experimental applications, p38γ-targeted research uses genetic deletion, kinase assays, and inhibitor-based pathway interrogation rather than broad p38α-centered assumptions[3][4][7].
